چکیده

AIMS To investigate the role of the PFC in the micturition reflex using an in vivo microdialysis study in rats. METHODS Adult female Sprague-Dawley rats were used and microdyalysis in the PFC and cystometrography (CMG) were performed under consciousness and free movement in the present study. Experiment 1: Samples including extracellular neurotransmitters were collected by microdyalysis and analyzed by high performance liquid chromatography. At the same time, CMG were performed to measure intercontraction interval (ICI) and maximum voiding pressure (MVP).  Experiment 2: SSRI (citalopram, 1 µM) was administered into the PFC, and microdyalysis and cystometrography (CMG) were performed simultaneously. Experiment 3: Following SSRI administration, 5-HT1A agonist (8-OH-DPAT, 300 µM), which has the effect of decreasing the level of serotonin (5-HT) in the PFC, was administered into the PFC, and microdyalysis and CMG were performed simultaneously. RESULTS Experiment 1: Extracellular level of 5-HT in the PFC significantly increased during micturition reflex (P < 0.05), whereas levels of glutamate or dopamine were not significantly changed. Experiment 2: Local administration of SSRI in the PFC increased the 5-HT level up to approximate 600% of the basal level. It also significantly increased ICI (P < 0.05), whereas no significant change was found in MVP. Experiment 3: The extracellular level of 5-HT gradually decreased after local administration of 5-HT1A agonist, thereby ICI significantly decreased (P < 0.05). CONCLUSIONS The results of the present study suggest that the PFC has a suppressive effect on neural control of the micturition reflex via serotonin. Neurourol. Effect of intracerebroventricular injection of GABA receptors antagonists on morphine-induced changes in GABA and GLU transmission within the mPFC: an in vivo microdialysis study

Objective(s): Many studies have focused on ventral tegmental area than of other mesocorticolimbic areas, and implicated a key role for the medial prefrontal cortex (mPFC) in the development of addictive behaviors. So far, the role of gamma-aminobutyric acid (GABA) receptors in the discriminative properties of morphine has received little attention and few studies evaluated the role of these rec...

Can ovariectomy and learning affect prefrontal cortex GABAAα1 receptor distribution in passive avoidance model in rats?

Introduction: The interaction between steroid hormones and neurotransmitters such as GABA has been proved. The regulation of muscimol binding to high-affinity GABAA receptors by estradiol and progesterone has been studied within distinct brain regions using in vitro quantitative autoradiography. There are few studies about the mechanism of the effect of steroid hormones on behaviors such as ...

Cinnamaldehyde improves methamphetamine-induced spatial learning and memory deficits and restores ERK signaling in the rat prefrontal cortex

Objective(s): Methamphetamine is a stimulant compound that penetrates readily into the central nervous system. Repeated exposure to methamphetamine leads to damage in the dopaminergic and serotonergic axons of selected brain regions. Previous studies showed that cinnamaldehyde improved memory impairment in animals. In the present study, we aimed to elucidate the effects of cinnamaldehyde on met...

Activity of the serotonergic system during isoflurane anesthesia.

BACKGROUND Microdialysis studies have demonstrated that the release of serotonin (5-hydroxytryptamine, 5-HT) in the serotonergic projection areas increases during waking and decreases during sleep in rat and cat, suggesting that 5-HT plays an important role in modulation of sleep. Although it might be expected that 5-HT release is also decreased during general anesthesia, the functional contrib...

Determination of 5-Hydroxymethyl-2-furaldehyde of Crude and Processed Fructus Corni in Freely Moving Rats Using In Vivo Microdialysis Sampling and Liquid Chromatography

The purpose of this study was to develop a sensitive and fast microdialysis coupled with high-performance liquid chromatographic (HPLC) method for determination of 5-hydroxymethyl-2-furaldehyde (5-HMF) in free-moving rats after i.g. administration of the aqueous extract of crud Fructus corni and its processed products of jiuzheng pin (JZP). The concentration of 5-HMF in free-movi...
